MGN-3 has been shown to be a powerful biological response modifier (BRM) as manifested by remarkable increase in natural killer (NK) cell activity in Immunocompromised individuals.
In the present study, we evaluated the immunomodulatory function of MGN-3 in 10 patients with different types of advanced malignancies.
Patients were under conventional therapy before participating in the study. Patients received MGN-3 at concentration of 45mg/kg/d orally for 2-6 months. Parameters under investigation included: NK cell activity against K562 tumor target cells. NK cell subpopulations using flow cytometry; and T and B cell functions. Patients were examined for these parameters at 0 day (baseline) and at 2 wks post treatment with MGN-3. Patients showed significant increase in NK activity ( more 2 fold) at 2 wks as compared to baseline value, while there was no change in NK cell populations. Patients also demonstrated elevation of T and B cell function as demonstrated by their proliferation response to different mitogens. The mechanism by which MGN-3 exerts its augmentory effect was examined and appears to be due to the production of cytokines such as TNF-α and IFN-Υ. We conclude that MGN-3 is a potent BRM and in the absence of notable side effects, MGN-3 may be useful in the immunotherapy of cancer. |
